摘要 A peptide linker comprising a novel thrombin cleavage site is provided to be useful for the recombinant production of fusion proteins comprising a desired protein and useful for separating the desired protein from the fusion protein. A peptide linker comprises a sequence of: Xl-X2-Ser-Pro-X3-X4-X5, wherein X1 is at least two amino acid residues which are the same or different from each other; X2 is a hydrophobic amino acid residue; X3 is arginine or lysine; X4 is alanine or glycine; and X5 is a non-acidic amino acid. A fusion protein comprises a desired protein, a fusion partner, and the peptide linker interposed therebetween, wherein the desired protein is selected from the group consisting of interleukin(IL)-11, thymosin beta4, thymosin alphal, 1L-2, IL-3, IL-4, IL-5, TL-6, IL-7, IL-8, IL-IO, IL-13, IL-15, IL- 18, protease-activated receptor 1(PARl), PAR3, PAR4, RANTES, stromal cell-derived factor-lalpha, monocyte chemotactic protein, stem cell factor, FLT-3L, parathyroid hormone, thrombopoietin, epidermal growth factor, basic fibroblast growth factor, insulin-like growth factor, granulocyte-macrophage colony stimulating factor, granulocyte colony stimulating factor, macrophage colony stimulating factor, platelet-derived growth factor, transforming growth factor(TGF)-betal, tumor necrosis factor(TNF)-alpha, interferon(IFN)-alpha, IFN-beta, IFN-gamma, hepatocyte growth factor, vascular endothelial growth factor and immunoglobulin heavy chain. A method for separating the desired protein from the fusion protein comprises a step of contacting the fusion protein with a sufficient amount of thrombin so that the peptide linker is cleaved.
