An simulated moving bed chromatographic separation device (SMB device), comprising an endless flow passage (2) formed with four columns (1a) to (1d) and connecting flow passages (2a) to (2d) connecting the columns (1a) to (1d) in series, and third and fourth flow passages (5) and (6) for discharging a mobile phase from any position of the endless flow passage (2). Regulators (5d) and (6d) and flow control valves (5e) and (6e) as flow rate adjusters for opening and closing main pipes (5a) and (6a) on the downstream side of the regulators (5d) and (6d) are provided each of in the third and fourth flow passages (5) and (6). Thus, the discharge of the mobile phase from the endless flow passage (2) can be controlled by a simple and highly durable structure.
