A method for activating pathway of bone formation is provided to increase activity and stability of Runx2 protein by inhibiting degradation of Runx2 protein by ubiquitylation through acetylation by BMP(bone morphogenic protein), so that it is useful for prevention and treatment of bone diseases including osteoporosis, osteogenesis imperfecta, periodontal disease and bone fracture. The transcription activity of Runx2 is increased by acetylation and stabilization of Runx2 protein by using BMP-2, p300, ubiquitylation of Runx2 by using Smurf1 or inhibiting deacetylation of Runx2 protein, wherein the acetylated region of Runx2 protein is lysine in the amino acid sequence of SEQ ID NO:1; and the deacetylation is carried out by histone deacetylase. The bone formation is increased by increasing Runx2 protein activity through acetylation, deacetylation inhibition and promotion of osteoblast differentiation.
