RECOMBINANT PLASMID, E.COLI TRANSFORMED THEREWITH AND METHOD FOR PRODUCING UBIQUITINATED PROTEIN BY USING THE SAME

PROBLEM TO BE SOLVED: To provide a recombinant plasmid for simultaneously expressing ubiquitin and a target protein in E.coli and ubiquitinate both proteins in the E.coli, an E.coli transformed with the recombinant plasmid, a method for screening (identifying and detecting) an unknown target protein by using the transformed E.coli and a method for producing and purifying a ubiquitinated or poly-ubiquitinated target protein.

SOLUTION: A target protein is ubiquitinated in E.coli by using: (A) an enzyme (E1) activating ubiquitin; (B) a binding enzyme (E2) for ubiquitin and a target protein; (C) a ligase (E3) for ubiquitin and a target protein; and (D) a recombinant plasmid capable of expressing ubiquitin.
