An operating method of a TPMS(Tire Pressure Monitoring System) is provided to prevent malfunction of the TPMS by enabling a TPMS receiver to receive signals transmitted from tire pressure sensors mounted on respective wheels and to check whether a tire pressure is suitable. An operating method of a TPMS equipped with a TPMS receiver comprise the steps of: receiving information on a tire pressure and so on detected from pressure sensors of respective tires(S1), detecting reception strength from information on signals when the signals are received from the sensors mounted on the respective wheels(S2), calculating a deviation from the detected reception strength(S3), determining discrimination of the calculated deviation(S4), distinguishing a rotating wheel and a spare wheel according to the deviation on the basis of a predetermined limit value, in the case of approval of the deviation(S5), and operating the TPMS on the basis of the rotating wheel, without regard for the spare wheel(S6).
