PROBLEM TO BE SOLVED: To provide a method for simply and certainly analyzing the site participated in the binding on a cell membrane surface substance. SOLUTION: The method of analyzing the binding site of the cell membrane surface substance is characterized in that (a) a native cell membrane surface substance and a cell membrane surface substance wherein the candidate binding site is modified is developed on the surface of a cell membrane, (b) the development amount of the cell membrane surface substance is investigated, (c) the binding amount of the binding substance to the developed cell membrane surface substance is investigated and (d) the binding ratio is calculated by dividing the binding amount by the development amount with respect to a native cell membrane surface substance and a cell membrane surface substance wherein the candidate binding site is modified, and the candidate binding site is determined to be participated in binding in the case where the binding ratio is changed by differentiating the candidate binding site.
