A DME(dimethyl ether)/diesel bi-fuel supply system of an in-line injection type diesel engine and a method for operating the same are provided to collect fuel, which is leaked from a fuel injection pump when DME fuel is used, and fuel, which is leaked from the fuel injection pump and an injector after the operation of the system stops. A DME/diesel bi-fuel supply system of an in-line injection type diesel engine comprises a DME fuel supply system(100) and a diesel fuel supply system(200). The DME fuel supply system(100) comprises a DME fuel tank(10), a fuel filter(13), a fuel supply pump(14), a fuel supply valve(15), a fuel injection pump(16), a plurality of injectors(17), an injector return pipe(18), a fuel injection pump return pipe(20), a fuel leakage pipe(21), an engine intake manifold(27), a purge tank(22), a purge pump(23), a purge valve(24) and a fuel purge pipe(26). The diesel fuel supply system(200) comprises a diesel fuel tank(30), a diesel fuel supply pump(32), a diesel fuel filter(35), a diesel fuel supply valve(36), a diesel injector return pipe(37), a diesel fuel injection pump return pipe(38), and a diesel collection pipe(39).
