PROBLEM TO BE SOLVED: To provide an improved quick method for determining the optimum concentration of a protective additive, without requiring preservation and analysis thereafter. SOLUTION: The method determines the optimum concentration of the additive capable of protecting sensitive biomacromolecules, for example, proteins or nucleic acids, against denaturation and aggregation in a solution including at least one species of a biomacromolecule and at least one species of a protective additive, comprising: (a) treating the solution with an ultrasonic wave; and (b) recording the value of one or more ultrasonic parameters of the ultrasonic velocity in the solution and/or the ultrasonic absorption of the solution. The method allows to detect a critical change of the ultrasonic parameter dependent on the concentration of the protective additive, and the change shows an interaction or a change of the interaction between the additive and biomacromolecules and/or between different biomacromolecules and/or a change of the conformation of the biomacromolecules. Thus, measurement of such changes can be used for determining the optimum concentration of the additive for protecting the biomacromolecule from denaturation and aggregation.
