摘要 <p>The method for investigating and determining protein containing biological material e.g. mites and pollen in a sample in regards to the symptoms of atopic diseases using Redprot (RTM: Pyrogallol-red dye) as protein detector, comprises applying test sample coated with the dye on a reflecting carrier material and investigating the biological materials with respect to structure and color tint. One square centimeter (cm2>) of the collected material or the test sample is coated with 0.2 ml of the Redprot (RTM: Pyrogallol-red dye) solution for three minutes. The method for investigating and determining protein containing biological material e.g. mites and pollen in a sample in regards to the symptoms of atopic diseases using Redprot (RTM: Pyrogallol-red dye) as protein detector, comprises applying test sample coated with the dye on a reflecting carrier material and investigating the biological materials with respect to structure and color tint. One square centimeter (cm2>) of the collected material or the test sample is coated with 0.2 ml of the Redprot (RTM: Pyrogallol-red dye) solution (0.02% solution pyrogallol-red in 30% isopropanol with pH 2.0) for three minutes. After the coloration, the remaining color solution is carefully removed and the sample is dried for two minutes at room temperature. For the sealing and preservation of the sample, a colorless medium of glycerine-gelatine-phenol is used, which is liquefied previously at 46 [deg]C, and then the sample for analysis is covered with a cover glass. The liquefied gelatine-glycerine-phenol (2 ml) is mixed with the Redprot (RTM: Pyrogallol-red dye) solution (1 ml) in a ratio 3:1. The pH value of the obtained or mixed solution is adjusted between 3.0-3.5 and the color solution is used immediately. The duration of coloring fluctuates between 4 and 6 hours. The analysis and image analysis of the sample takes place using A CCD camera or video, a spectral analysis in the spectrophotometer, fluorescence analysis in fluorescence microscope or spectral analysis in confocal laser scanning microscope.</p>
