PROBLEM TO BE SOLVED: To provide a method for preparing a sample for detection by which an analyzing time can be shortened by simplifying a pretreatment method of a biosample usable for detecting the methylation of a DNA, and further to provide a method for detecting the methylation of the DNA, a sample-treating liquid and a kit. SOLUTION: The method for preparing the sample for the detection usable for detecting the methylation of the DNA includes a step for mixing a biosample containing a cell with the sample-treating liquid containing a surfactant for solubilizing the cell, and a step for adding a converting agent for converting an unmethylated cytosine contained in the DNA to other bases.
