A METHOD TO EXPRESSION AND PURIFICATION OF XOR I, THE ENDONUCLEASE OF XANTHOMONAS ORYZAE PV. ORYZAE

An endonuclease Xor I of Xanthomonas oryzae pv. oryzae, Xoo is provided to replace Pst I, widely and commercially used isoschizomer, due to having excellent characteristics. And a method for preparing a recombinant protein Xor I is provided to mass-produce the recombinant protein Xor I at relatively low cost. An Xor I endonuclease derived from Xanthomonas oryzae pv. oryzae, Xoo is described as SEQ ID : NO. 1. A method for mass-producing a recombinant protein Xor I comprises the steps of: (a) culturing a transformant to which a recombinant expression vector including an Xor I gene is introduced to express an Xor I protein; (b) after centrifuging the strain culture solution including the expressed protein to obtain a pellet, suspending it in a buffer solution; (c) performing chromatography using the buffer solution of the step (b); and (d) eluting a chromatography-performed column of the step (c).
