PROTEIN EXPRESSION IN BACULOVIRUS VECTOR EXPRESSION SYSTEM

PROBLEM TO BE SOLVED: To provide a method for producing vaccines for increasing protein antigen expression in baculovirus vector expression systems.

SOLUTION: A method containing following steps and for producing vaccine containing classical swine fever virus E2 baculovirus protein is provided. Wherein the steps are a step for selecting a recombinant baculovirus encoding the protein, a step for growing insect cells in a growth medium in a culture vessel with a sufficient volume of at least 2 liters, a step for infecting the cells with an inoculum of the baculovirus at 1×105to 5×106cells/mL cell density with <0.01 MOI, a step for culturing the cells till more than about half of the cells exhibits cell modification effects and removing cellulous materials when a final concentration of the E2 protein in the growth medium is 100-300μg/mL, and a step for preparing the vaccine without immunoaffinity purification. And furthermore the method can be applied to produce recombinant follicle stimulating hormone.
