PROBLEM TO BE SOLVED: To provide a method for measuring calcium ion, usingα-amylase activity, wherein the method can accurately and simply measure calcium ion in a body fluid widely from low values to high values, and to provide a reagent composition for measurement and usable for the method. SOLUTION: The reagent composition for measuring calcium ion in the body fluid is provided, wherein the composition contains inactiveα-amylase, 2-chloro-4-nitrophenyl 4-O-β-D-galactopyranosyl-α-maltoside, if required, maltooligosaccharide or the maltooligosaccharide having reduction terminal glucose bonded with a non-chromophore, and sodium chloride, and the final pH of the composition is kept between 5.5-6.5.
