PROBLEM TO BE SOLVED: To provide a noncontact detection means, capable of determining on the spot whether a fine monocrystal of protein molecules generated in a sample solution drop is a monocrystal, having superior crystallinity or a monocrystal having crystallographic orientation disturbance in the protein molecules, when utilizing a hanging drop-vapor diffusion method. SOLUTION: In the fine monocrystal of the protein molecules generated in the sample solution drop, concerning the Raman scattered intensity of the Stokes line caused by vibration modes: amide I, amide III derived from an amide bond (-CO-NH-) originated inα-helix structure contained in the protein molecule, the dependence on angleθ, formed between the polarization direction of linearly-polarizing incident laser light, the spiral axial direction ofα-helix is measured by polarized Raman scattering, and the degree of the crystallographic orientation disturbance in the protein molecules is determined, based on the magnitude of angle dispersion.
