A method for improving transfection efficiency by synchronization of a cell cycle in cells is provided to maximize the transfection efficiency of mammal cells by making many cells be the status to receive foreign DNA effectively. A method for improving transfection efficiency comprises the steps of: (a) culturing a mammal cell in a medium containing 0-0.5% of serum for 12-36 hours; (b) performing transfection during the culture through lipofection using cationic lipid; (c) replacing the culture medium by the normal medium containing serum after 3-12 hours of transfection; (d) culturing the mammal cell for 12-48 hours after medium replacement; (e) removing the medium after culture; and (f) collecting the cells after medium removal to prepare cell eluate, wherein the mammal cell is a normal cell selected from Chang(HeLa, liver tissue, human), 293(Hek293, embryonal kidney, human), L929(connective tissue, mouse), NIH/3T3(embryo, mouse), PC-12(adrenal gland, rat), CHO(ovary Chinese hamster) and Cos7(kidney, monkey).
