TRANSFORMANT EXPRESSING NITRILE HYDRATASE

A novel microorganism is provided to express nitrile hydratase showing high stability on heat or highly concentrated nitrile and amide compounds, thereby capable of efficiently converting the nitrile compound into the corresponding compound. The Rhodococcus microorganism is transformed by DNA described by either (A) or (B), wherein the Rhodococcus microorganism is Rhodococcus rhodochrous M33 strain(VKM Ac-1515D or KCCM-10635) and/ or a mutant thereof. The DNA (A) comprises an amino acid sequence described at a SEQ ID : NO. 1 of a sequence list corresponding to an alpha sub-unit of nitrile hydratase or a sequence coding an amino acid sequence having at least one substitution, deletion or addition of amino acid in the amino acid sequence described at the SEQ ID : NO. 1 of the sequence list, and an amino acid sequence described at a SEQ ID : NO. 2 of the sequence list corresponding to a beta sub-unit of the nitrile hydratase or a sequence coding an amino acid sequence having at least one substitution, deletion or addition of amino acid in the amino acid sequence described at the SEQ ID : NO. 2 of the sequence list; and codes a protein having the nitrile hydratase activity. The DNA (B) comprises a sequence from 695th to 1312th of a SEQ ID : NO. 3 of the sequence list corresponding to a sequence coding the alpha sub-unit of the nitrile hydratase or a sequence hybridizing the corresponding sequence under strict condition, a sequence from 1st to 681st of the SEQ ID : NO. 3 of the sequence list corresponding to a sequence coding the beta sub-unit of the nitrile hydratase or a sequence hybridizing the corresponding sequence under strict condition; and codes a protein having the nitrile hydratase activity.
