PROBLEM TO BE SOLVED: To provide an inexpensive method for separating and purifying a nucleic acid which can recover a high yield of RNA from a mixture sample of the RNA with DNA, excellent in separation performance, having high cleaning efficiency, simple, rapid and excellent in suitability for automatization. SOLUTION: The method for separating and purifying the RNA comprises (1) a process for preparing a mixture solution by adding a nucleic acid solubilization reagent to a biomaterial and preparing a sample solution by adding a water-soluble organic solvent or a solution containing the water-soluble organic solvent to the mixture solution, (2) a process for adsorbing the nucleic acid to a nucleic acid-adsorbing porous membrane by passing the sample solution containing the nucleic acid through the membrane, (3) a process for cleaning the nucleic acid-adsorbing membrane in an RNA adsorbed state by passing a cleaning solution through the porous membrane and (4) a process for releasing the RNA from the nucleic acid-adsorbing membrane by passing a recovery solution through the porous membrane. In the method, the nucleic acid-adsorbing porous membrane is a porous membrane which adsorbs the nucleic acid with mutual actions to which ionic bonds are not participating and the sample solution contains the water-soluble organic solvent in the concentration of 10-35 volume%.
