Measles vaccine is prepared by successively cultivating measles, virus obtained from a human source, in at least five primate cell tissue cultures, each culture comprising the same type of cells, and processing the resulting virus into a vaccine in a conventional manner. Suitable cell cultures are those of monkey or human embryo tissue, e.g. heart, muscle, testicles, skin, spleen, tonsils, thyroid, annion or chorion, or human diploid cells. Up to twenty successive cultivations may be employed. The cultivated virus is processed into a vaccine by inactivation with formaldehyde or treatment with ether and a non-ionic surfactant, e.g. polyoxyethylenesorbitan mono-oleate or mono-stearate, alkyl-phenol polyglycol ethers, ethylene oxide-higher alcohol or -fatty acid condensates,. or sodium desoxycholate. When processed with such a surfactant, the measles vaccine may be combined with pertussis antigens and additionally with other vaccines, eg. diptheria and/or poliomyelitis and/or tetanus. An adjuvant, such as aluminium hydroxide or phosphate, is preferably added to the measles vaccine or combined vaccine. '</p> |
