Improved kanamycin kinase II (I), or a variant or functional fragment, that includes at least one of the substitutions Gln155Trp and Ser114His is new. Independent claims are included for the following: (1) fusion protein (FP) of (I) and a polypeptide of interest (II); (2) nucleic acid (III) that encodes (I) or FP; (3) expression cassette (EC) for (III); (4) recombinant vector that contains (III) or the cassette of (3); (5) method for selecting a cell transformed or transfected by a recombinant vector that contains (III) or EC; (6) method for preparing a transformed plant that includes EC or the vector of (4); (7) library of variants of (II) comprising a polynucleotide that encodes a variant of a (II) fused in phase with (III); and (8) method for producing soluble variants of a protein/peptide of interest that comprises generating a bank of mutant genes encoding a protein/peptide of interest fused to (III).
