摘要 PROBLEM TO BE SOLVED: To provide a method for detecting a target nucleic acid having a stem-loop structure by hybridizing it with a probe nucleic acid in improved hybridization efficiency. SOLUTION: The method for detecting a target nucleic acid having a stem-loop structure is provided, comprising the following procedure: Part of the loop structure portion of the target nucleic acid is defined as a to-be-detected sequence, a probe nucleic acid having a sequence complementary to the to-be-detected sequence and a protective nucleic acid having a sequence complementary to the sequence in the portion other than that of the to-be-detected sequence are prepared, and the protective nucleic acid is bound to the target nucleic acid to make a hybridization between the target nucleic acid and the probe nucleic acid, thus detecting the target nucleic acid bound to the probe nucleic acid.
