PROBLEM TO BE SOLVED: To provide a therapeutic composition containing a preparation useful for the treatment of TNF-mediated pathology. SOLUTION: The invention relates to a method for controlling the amount of sialic acid on an oligosaccharide side-chain of a mammalian glycoprotein produced by mammalian host cell culture. The method containing the culture of a mammalian host cell in the production phase of the cultivation is characterized by (i) the addition of a 3-6C alkanoic acid or its salt in the cell culture system at a concentration of 0.1-20 mM, (ii) the maintenance of the weight osmol concentration to 250-600 mOsm in the cell culture and (iii) the maintenance of the culture temperature to 30-35°C.
