The present invention relates to MCP-1 muteins, including specified portions or variants thereof, which may, optionally, be PEGylated or biotin-PEGylated. The invention further relates to chemical synthetic methods for producing the MCP-1 muteins and corresponding conjugates. The invention relates to the use of said muteins and particularly to the use of the biotinylated conjugates in selection of neutralizing antibodies to human MCP-1 protein and the antibodies so selected. The antibodies of the invention are specific for an epitope conserved on native MCP-1 conserved in the at least one biotinylated and PEGylated MCP-1 mutein protein or fragment thereof.
