摘要 Signal amplification methods for measuring a very small amount of protein by using mass spectrometry are provided to improve precision of the protein amount measurement by indirectly amplifying the protein and measuring the amplified signal with the mass spectrometry. The signal amplification method for measuring the very small amount of protein comprises the steps of: (a) linking a specific enzyme to an antibody so as to prepare an antibody conjugate; (b) reacting the antibody conjugate with an immobilized or non-immobilized protein; (c) adding a substrate into the complex; and (d) measuring the reaction product by using the mass spectrometry such as nano-LC(liquid chromatography)/MS(mass spectrometry)/MS or MALDI-TOF(matrix-assisted laser desorption ionization time of flight) mass spectrometry, wherein the enzyme is linked to a Fab(fragment-antigen binding) domain of the antibody, and an inactive enzyme or an enzyme capable of activating the inactive enzyme.
