A coupling structure of an engine and an automatic transmission in a vehicle is provided to reduce a manufacturing cost by simplifying design conditions of a drive plate and securing durability of the drive plate. A coupling structure of an engine and an automatic transmission in a vehicle is used for connecting a crankshaft of the engine with a torque converter(20) of the automatic transmission. A vertical long hole(15) is formed in a drive plate(10). A flange-integrated stud(25) is formed at the torque converter in order to be combined with the vertical long hole of the drive plate. The length of the stud is determined according to a movement toward an axial direction in order not to penetrate the vertical long hole of the drive plate.
