PROBLEM TO BE SOLVED: To perform the cloning and purification of a thermostable DNA polymerase derived from Bacillus stearothermophilus. SOLUTION: A truncated thermostable Bst DNA polymerase is produced by eliminating 5'-3' exonuclease activity from a Bst DNA polymerase using recombinant DNA and protein fusion techniques.
