A steering angle sensor automatic setting system of a vehicle and a method thereof are provided not to occur a tolerance by performing an adjustment of a zero point about a steering angle sensor at a running test mode for a practical vehicle after being fully assembled. A method of a steering angle sensor automatic setting system of a vehicle includes the steps of monitoring a velocity of vehicle(S101), checking whether the velocity of vehicle exceeds a standard velocity or not(S102), measuring a length acceleration velocity and a width acceleration velocity of vehicle(S103), checking a measuring value is zero or not(S104), comparing wheel velocities about four wheels(S105), checking whether the four wheel velocities are equal or not(S106), completing a command control of a steering angle sensor(S107), increasing an implementation count(S108), checking the set number(S109), storing an average of zero point controlling value as a rotation value(S110), turning off a VDC(Vehicle Dynamic Control system) alarm light(S111), and completing the zero point adjustment of the steering angle sensor(S112).
