A transdermal delivery composition comprising non-steroidal anti-inflammatory drugs(NSAID) and local anesthetics is provided to increase skin permeation and enhance medicinal effects of the anti-inflammatory drugs by improving solubility of non-steroidal anti-inflammatory drugs, and reduce skin irritation by using local anesthetics of non-chloride. The transdermal delivery composition comprises 0.1-30 wt.% of non-steroidal anti-inflammatory drugs, 1-20 wt.% of local anesthetics of non-chloride, 30-80 wt.% of sticky polymer and 0.1-20 wt.% of transdermal absorption-promoting agent, wherein the transdermal absorption-promoting agent is oleic acid, oleyl macrogol-6-glyceride, lauroylsarcosine, isopropyl myristate, capryl caproyl macrogol-8-glyceride, polyglyceryl-3-isostearate, polyglyceryl-6-diolate, polyoxyethylene-10-oleylether, propyleneglycol monolaurate or propyleneglycol monocaprate; the non-steroidal anti-inflammatory drug is ibuprofen, ketoprofen, flurbiprofen, feno-profen, naproxen, piroxicam, tenoxicam, isoxicam, melo-xicam, indomethacin, aceclofenac or diclo-fenac; the local anesthetic is procaine, chlorocaine, tetracaine, prilocaine, lidocaine, mepivacaine, bupivacaine, ropivacaine or etidocaine; and the sticky polymer is acryl adhesive of acrylate polymer or vinylacetate acrylate copolymer, or synthetic or natural rubber and silicon-based adhesive selected from polyisobutylene, polystyrene, polybutadiene and copolymer thereof.
