PROBLEM TO BE SOLVED: To provide a method capable of detecting clearly an antigen-antibody reaction (the presence thereof and a degree thereof), while using an atomic force microscope such as an atomic force microscope. SOLUTION: In this method, an antibody is immobilized in one of a probe and a substrate in the atomic force microscope, an antigen is immobilized in the other thereof, the probe is once brought into contact with the substrate in a solution, and force required when separating gradually the probe from the substrate is measured thereafter to detect the antigen-antibody reaction. The method uses a solution containing a degradation product of one kind of substance selected from a protein, a peptide, and a long chain hydrocarbon compound having a sugar chain or hydrophilic group, as the solution used for the measurement.
