PROBLEM TO BE SOLVED: To provide a measuring method applicable to an automatic analyzer, capable of accurately, quickly and simply measuring with excellent reproducibility, an interaction between a biomolecule and a molecule interacting with calcium ions and having a structure changed according to the concentration of the calcium ions. SOLUTION: This measuring method of the interaction between the biomolecule and the molecule interacting with calcium ion, and having a structure changed according to the concentration of the calcium ions is characterized by including: a process for performing fluorescence labeling of the molecule interacting with calcium ions, having a structure changed according to the concentration of the calcium ions; a process for mixing the fluorescence-labeled molecule with the biomolecule; a process for measuring fluctuation of the fluorescence; and a process for detecting the interaction between the biomolecule and the molecule having a changing structure by analyzing a measurement result of the fluctuation of the fluorescence.
