<P>PROBLEM TO BE SOLVED: To provide a new method for amplifying a nucleic acid by which the specificity for a nucleic acid sequence to be a target is enhanced. <P>SOLUTION: The method for amplifying the nucleic acid comprises a step of binding a first primer complementary to a first sequence and a second primer complementary to a second sequence to the target nucleic acid having the first sequence, the second sequence and a third sequence existing between the first and the second sequences, a second step of synthesizing a first amplified double-stranded nucleic acid having the third sequence and a prescribed sequence, a third step of carrying out an exonuclease treatment for the resultant product of the second step, a fourth step of separating the first amplified double-stranded nucleic acid into a single-stranded nucleic acid with the prescribed sequence, a fifth step of binding a third primer complementary to the prescribed sequence and a fourth primer complementary to the third sequence to the single-stranded nucleic acid with the prescribed sequence and a sixth step of synthesizing a second amplified double-stranded nucleic acid. <P>COPYRIGHT: (C)2007,JPO&INPIT |
