Diagnosis and control of progression of sepsis or general acute inflammation is performed in vitrio, by measurement of concentration of extra-cellular hemoglobin, and/or its sub-components, and particularly the beta chain or its sub-components and/or its derivatives. Diagnosis is made by comparision of the measured concentration against results obtained from a control group. The procedure may be performed either as a laboratory chromatographic or 'on-micro chip' determination (claimed). ACCP/ SCCM ( American College of Chest Physicians/Society of Critical Care Medicine) Consensus Conference 1992, defined 'SIRS (systemic inflammatory response syndrome)' and and 'Sepsis'. An example describes ammonium sulphate precipitation of protein from plasma samples, and chromatographic removal of immunoglobuline (IgC) and albumin from the solution in sodium phosphate, on a G-Separose column, followed by fractionation on anionic exchange resin. Buffer eluted fractions were analysed by 2D electrophoresis on polyacrylate gel, and spots developed with silver. MALDI Peptide mass fingerprint technique was used for mass spectrometric comparision of spot intensities between patient and control samples. A comprehensive reference bibliography is included in the specification.
|
