MUTATED FORMS OF NY-ESO-1 PRODUCED IN YEAST AND USES THEREOF

Highly immunogenic protein NY-ESO-I is produced poorly in recombinant eukaryotic system. "Directed evolution" and improved purification techniques resulted in a mutein of NY-ESO-I, wherein a cysteine culture at positions 75-78 (CCRA), is replaced by AARA and position 153 is mutated from leucine to histidine. The mutein is obtained easily from yeast and is as immunogenic as wild type NY-ESO-I in MHC-Class I, HLA-A2<SUP>+</SUP> systems.
