<p>Methods are provided for rapid amplification of large DNA including genome amplification. The method utilizes nickases and polymerases but does not utilize primers. Amplification of a large DNA can be achieved within about 10 minutes as determined by detectable bands on gels starting with 25 ng DNA in a volume of 50 µl. An advantage of these methods of amplification for large DNA is that the sequence is proportionally represented. The methods can further provide amplified DNA in a time-efficient manner. The nick- based amplification can be supplemented by a primer-based amplification for increasing the concentration of particular target sequences. The methods can also be used to provide a fingerprint profile of the DNA.</p> |
