<P>PROBLEM TO BE SOLVED: To provide a nucleic acid for producing a short hairpin RNA (shRNA) precursor, and utilization thereof. <P>SOLUTION: The nucleic acid for producing the shRNA precursor contains the following components (1) and (2): (1) an RNA polymerase type II promoter; and (2) a shRNA precursor-encoding region comprising the following (i) to (iii): (i) a Drosha recognition first region containing a Drosha recognition site; (ii) a shRNA-forming region; and (iii) a Drosha recognition second region containing the Drosha recognition site, wherein the shRNA-forming region of 2-(ii) comprises (a) a sense strand region encoding a sense sequence which is a base sequence homologous to at least a part of a base sequence of a target gene which can be a target of RNA interference, (b) a loop region encoding a loop sequence to be cleaved by a base-specific RNase, and (c) an antisense region encoding an antisense sequence complementary to a sense sequence which is a base sequence homologous to at least a part of a base sequence of the target gene. <P>COPYRIGHT: (C)2007,JPO&INPIT |
