Disclosed are: a method for modification of the C-terminus of a protein or peptide at low cost, in a simple manner and with good efficiency; a method for isolation of a C-terminal peptide fragment of a protein or peptide in a simple and highly reliable manner; and a method for rapid sequencing of an amino acid sequence for a protein or peptide using a mass spectrometer with high accuracy and reliability. Specifically, disclosed are: a method for converting a carboxyl group in a protein or peptide into an aldehyde group by mixing the protein or peptide with a formulation reagent and a catalyst; a method for modifying the C-terminus of the protein or peptide by reacting the aldehyde group with a nucleophilic reagent; a method for immobilizing the protein or peptide by reacting the aldehyde group with a support having a nucleophilic group; a method for isolating a C-terminal peptide fragment from the support by digesting the immobilized protein or peptide and then washing the support; and a method for sequencing an amino acid sequence of the isolated C-terminal peptide fragment by subjecting the isolated C-terminal peptide fragment to mass spectrometry.
