This invention relates to a method of analyzing nucleic acid with regard to composition and/or sequence comprising (a) bringing a sample comprising said nucleic acid into contact with positively charged and/or charge-neutral groups, said groups being attached to a solid support and under conditions allowing binding of said nucleic acid to said groups; (b) performing one or more washing steps; (c) depositing MALDI matrix material onto said solid support which has been contacted with said nucleic acid in step (a); and (d) performing MALDI-MS of said nucleic acid.
