PROBLEM TO BE SOLVED: To provide a method for screening a substance effective for preventing and treating obesity ineffective by exercise therapy, with which a mouse having reduced AMPKα1 andα2 function, capable of screening a substance useful for preventing or treating obesity, is produced and used. SOLUTION: A variant mouse having reduced AMPKα1 andα2 function is produced. For example, a target substance for screening is administered to a mouse having reduced AMPK activity, the mouse is exercised in a load to reduce the fat amount of a normal mouse but not to significantly reduce the fat amount of the mouse in comparison with a normal mouse. Whether the fat amount is significantly reduced or not is examined by comparison with a case in which the substance is not administered to the mouse.
