A method for preparing 2,6-naphthalene dicarboxylic acid is provided to efficiently and economically produce highly purified 2,6-naphthalene dicarboxylic acid by biologically converting 2-formyl-6-naphthoic acid into the 2,6-naphthalene dicarboxylic acid using aldehyde dehydrogenase derived from Bacillus subtilis. The method comprises the steps of: (a) after mixing a buffer solution selected from the group consisting of water, KH2PO4-KOH, sodium pyrophosphate-HCl, boric acid-NaOH and sodium borate-HCl, dimethyl sulfoxide and an NAD solution with crude naphthalene dicarboxylic acid, adjusting the pH of the mixture solution to 7.5-8.5 so as to obtain a purification reaction solution; and (b) reacting an aldehyde dehydrogenase enzyme purification solution obtained by purifying a culture solution of microorganism, which is obtained by transforming the microorganism with a recombinant expression vector including an aldehyde dehydrogenase gene derived from Bacillus subtilis HSB-21(deposition no. KFCC-11211) described as SEQ ID : NO. 1, with the purification reaction solution obtained from the step(a) at a temperature of 30-45 deg.C so as to convert 2-formyl-6-naphthoic acid contained in the crude naphthalene dicarboxylic acid into 2,6-naphthalene dicarboxylic acid.
