摘要 PROBLEM TO BE SOLVED: To provide a method and a kit for detecting genetic polymorphism using a fluorescence proximity method with which general-purpose properties of detection equipment are high and detection in an open system can be carried out. SOLUTION: The method for detecting the genetic polymorphism is carried out as follows. A first oligonucleotide (having a sequence complementary to a third region, a sequence complementary to a second region, a non-complementary flap sequence and a fluorescence energy donor or receptor in the flap sequence) and a second oligonucleotide (having a sequence complementary to a first region and an optional sequence) are hybridized with a specific nucleic acid in a sample having a specific sequence (composed of the first region, the second region and the third region contiguous in order from the downstream side in which the second region is composed of a polymorphic base to be detected). Thereby, a triple-stranded structure is formed and a nucleic acid fragment containing the flap sequence is liberated with an FEN (flap endonuclease) to detect the fluorescence from the liberated nucleic acid fragment by using a third oligonucleotide (having a sequence complementary to the flap sequence and the fluorescence energy receptor or donor).
