PROBLEM TO BE SOLVED: To provide an inexpensive and highly sensitive method for determining DNA sequence without using an expensive reagent in a stepwise complementary chain-synthesizing reaction. SOLUTION: dATP in an amount not exceeding 50 times of the amount of a template DNA is added to a reactor containing the template DNA; stepwise complementary chain synthesis is carried out; background emission intensity originated from dATP in which dATP becomes a substrate of luciferase and emits light is deducted from the measured luminescence; and luminescence resulting from complementary chain synthesis caused by ATP converted and produced from pyrophosphoric acid produced in complementary chain synthesis is obtained.
