METHOD FOR PREPARING ESCHERICHIA COLI PHYTASE USING PROMOTER DERIVED FROM YEAST

A method for preparing phytase derived from Escherichia coli is provided to increase the expression of the phytase and obtain the phytase having high activity by transforming a recombinant expression vector prepared by operably linking a nucleic acid sequence encoding phytase derived from Escherichia coli to a promoter derived from yeast with Escherichia coli and then culturing the Escherichia coli. The recombinant expression vector comprises a nucleic acid sequence encoding phytase derived from Escherichia coli to be operably linked to a control sequence including a promoter derived from yeast such as GAL10 promoter. The method for preparing phytase derived from Escherichia coli comprises the steps of: (a) preparing a nucleic acid sequence encoding the phytase derived from Escherichia coli; (b) operably linking the nucleic acid sequence to a promoter derived from yeast to prepare a recombinant expression vector; (c) transforming Escherichia coli host cell line using the recombinant expression vector; (d) culturing the transformed Escherichia coli host cell line to express phytase; and (e) isolating the phytase derived from Escherichia coli from the culture product and then purifying it.
