<P>PROBLEM TO BE SOLVED: To provide an enzyme useful as a detergent enzyme acting in an alkaline region, having xyloglycan decomposition activity and activity to decompose crystalline cellulose and provide a gene for the enzyme. <P>SOLUTION: The alkaline xylanase has the following enzymatic properties. (1) Action: It hydrolyzes β-1,4-glycoside bond of xylan, (2) substrate specificity: it decomposes xylan and crystalline cellulose, especially xylan, (3) optimum reaction pH: the optimum reaction pH is about 8.5 in the case of using xylan as a substrate and reacting at 40°C, (4) pH stability: the enzyme treated at 5°C for 20 hr using xylan as a substrate has residual activity corresponding to ≥80% of the activity of untreated enzyme at pH 4.0-10.5 revealing high stability of the enzyme, (5) optimum reaction temperature: the optimum reaction temperature is about 50°C in the case of reacting at pH 9.0 for 20 min using xylan as a substrate, (6) thermal stability: the enzyme treated at pH 7.0 for 20 min using xylan as a substrate has residual activity corresponding to ≥90% of the activity of untreated enzyme at ≤50°C revealing high stability of the enzyme and (7) molecular weight: the molecular weight is about 40 kDa determined by SDS-polyacrylamide gel electrophoresis. <P>COPYRIGHT: (C)2007,JPO&INPIT |
