<p>The invention provides a method of making a mutated Type IIT endonuclease which has nicking activity comprising the steps of:
(a) identifying a heterodimeric Type IIT endonuclease;
(b) identifying a conserved region within said Type IIT endonuclease;
(c) generating at least one mutation within said conserved region; and
(d) analyzing the mutant endonuclease of step (c) for nicking endonuclease activity.</p>
