PROBLEM TO BE SOLVED: To measure a specific substrate in a sample, especially the concentration of glucose in humor, at a potential insusceptible to ascorbic acid and oxygen by employing an electrode containing a carbon electrode material, an oxidizing/reducing enzyme and a mediator compound, i.e., nickelocene or a derivative thereof. SOLUTION: A working electrode (biosensor) 12 containing a carbon electrode material, an oxidizing/reducing enzyme and a mediator compound, i.e., nickelocene or a derivative thereof, is immersed into a sample 26 to be measured in a container 24 disposed in a thermostatic bath 28 at about 30 deg.C along with a reference electrode (Ag/AgCl) 14 and a counter electrode (Pt) 16 and then the sample 26 is stirred by means of a stirrer. A potential difference in the range of -400mV to +200mV is then applied between the electrodes 12, 14 and the quantity of current (quantity of response current) flowing between the electrodes 14, 16 is measured. A specific substrate in the sample 26, especially the concentration of glucose in humor, is determined based on a working curve prepared for a standard sample for which the concentration of specific substrate is known. |
