PROBLEM TO BE SOLVED: To provide a simple method for producing a phosphoroamidite compound with its basic moiety nonprotected, with no need of any complicated reaction such as a hydroxyl-selective reaction. SOLUTION: The method comprises deprotecting the basic moiety of the corresponding basic moiety-protected phosphoroamidite compound with an ammonia/methanol solution or methylamine/THF solution. It is preferable that the basic moiety-protected phosphoroamidite compound be such that the basic moiety of a nucleoside is protected followed by protecting the 5'-OH group and then the 3'-OH group is phosphoroamidite-modified. Thereby, this simple method can be afforded with no need of any complicated reaction such as a hydroxyl-selective reaction, enabling the objective phosphoroamidite compound with its basic moiety nonprotected to be mass-produced.
