A fibrinolytic protease derived from Formitella fraxinea is provided to have the substrate specificity different from that of a conventional fibrinolytic enzyme, and show excellent fibrinolytic activity on fibrin and fibrinogen, thereby being effectively used for preventing and treating thrombus. The fibrinolytic protease derived from Formitella fraxinea is characterized in that it is a metalloprotease, has a molecular weight of about 42 kDa, shows an optimum temperature of 20-45 deg.C and an optimum active pH of 5.0-7.0, and is able to decompose fibrin or fibrinogen within 1 hour. The activity of fibrinolytic protease is deteriorated by metal ions such as Cu^2+, Fe^3+, and Zn^2+.
