A spherical digestive enzyme granule is provided to show high sphericity and grain size uniformity, and have excellent enzymatic stability and formulating advantages by coating an aqueous dispersion of a digestive enzyme on a spherical core. The spherical digestive enzyme granule comprises a spherical core and a digestive enzyme layer coated on the core. The method for preparing the granule comprises the steps of: (a) dissolving at least one digestive enzyme selected from the group consisting of protease, lipase, amylase, cellulase, hemicellulase, xylanase, beta-glucanase, lactase, alpha-galactosidase, phytase, phospholipase, beta-glucosidase, and nattokinase and a pharmaceutically acceptable carrier in a buffer solution which is pH controlled so as to maintain the optimum activity of the digestive enzyme; (b) mixing the dissolved enzyme with an organic solvent to prepare a digestive enzyme organic solvent aqueous dispersion; and (c) spraying the aqueous dispersion into a core to be absorbed thereinto and grow particles.
