A method for assaying protein is provided to be able to analyze extremely small amount of protein by amplifying a surface plasmon resonance(SPR) signal using a non-labeled antibody through a sandwich technique, thereby being variously applied to a disease detecting sensor, proteomics study and the like. The method comprises the steps of: (a) forming a self-assembled mono-molecule layer having carboxyl group and hydroxyl group on the surface of a metal thin film on a SPR chip, where the metal is selected from the group consisting of Au, Ag, Cu and Al; (b) biotining the self-assembled mono-molecule layer of the SPR chip and then combining it with avidin or streptavidin; (c) fixing the avidin or streptavidin of the SPR chip on a target protein using a specific and biotined primary antibody; (d) reacting the fixed antibody of the SPR chip with a biological sample such as tissue, cell, whole blood, serum, plasma, saliva, sputum, and urine including the target protein; (e) reacting the SPR chip with a specific and non-labeled secondary antibody; and (f) measuring the refractive index change of the SPR chip on the material fixed on the metal thin film surface to calculate the amount of the target protein.
