A cup holder of a vehicle is provided to safely support a cup by adding a separation preventing structure on the upper end of a cup guide and preventing the upper end of the guide from being separated even under excessively strong operating force in rotating the cup guide while settling the cup. A cup holder of a vehicle comprises a housing(10) formed with a guide line(11) at both sides to a certain length, a first spring mounting tool(12) protruded inward at the front side, and a supporting member(13) with a settling groove(14) at the upper side to have cylindrical grooves; a guide(20) having a separation preventing tool(22) hinge-combined with the settling groove, a second spring mounting tool installed correspondingly to the first spring mounting tool, and a moving shaft(23) moved forward and backward along the guide line; and a spring installed at the first and second spring mounting tools to elastically connect the housing with the guide. A groove is formed on the inner lower side of the guide to elongate a time point for contacting the guide with the housing.
