A sliding door opening/closing device of a vehicle is provided to automatically open/close a door usually by automatically and manually opening/closing a sliding door and to reduce the number of working processes and defective rate by making the structure compact. A sliding door opening/closing device of a vehicle comprises a frame(100) installed on a vehicle to have a transferring unit(10); a gear unit(20) installed at the frame to transfer the driving force by a driving motor(70); a link unit(30) rotatively combined with the transferring unit and the gear unit, and composed of a first link(31) connected to a driven gear(22) and a second link(32); a buffering means(40) installed between a sliding door and the link unit to absorb impact while opening/closing the sliding door; a manual/automatic adjusting unit(50) selectively operating the gear unit to manually and automatically open/close the sliding door; and a guide unit(80) installed at the frame to induce rotation of a second link(32).
